It is known, that cardiovascular (CVDs) diseases are the most common causes of death around the world. The main challenge to treat CVDs is the fact that the damaged cardiomyocytes (CMs) do not renew. However, the newest research indicates potential of stem cells (SCs) to regenerate heart tissue. To define proper parameters for cell regeneration, SC differentiation towards heart cells should be studied under conditions as close as possible to native tissue. Conventional culture models do not fulfill such requirements. Perfusion Lab-on-a-chip systems, mimicking heart tissue features, seem to be a suitable tool used for optimization parameters of heart cell regeneration. Although, Heart-on -a- chip systems used to cardiac cell culture and study drug cardiotoxicity have been developed, some additional features of heart (which enhance cardiac cell maturation) are often skipped.

The goal of the proposed Project is to study how parameters such as perfusion, nanofibers, stem cells and biochemical factors influence regeneration of heart cells under disease (hypoxia) state. For this purpose, a new Lab-on-a-chip system will be developed and integrated with additional biomaterials (a new fabricated nanofibrous mats), which should enhance cardiac cell maturation and stem cells differentiation towards cardiac cells. The geometry of the microsystem will be optimized and designed in such a way that both mono- and cocultures of cardiac and stem cells will be performed simultaneously.
